化香树果序没食子酸和鞣花酸含量测定及HPLC指纹图谱研究

摘要:

目的采用HPLC方法同时测定化香树果序没食子酸和鞣花酸并对12批化香树果序药材进行指纹图谱研究。方法采用Acclaim TM 120A C18色谱柱(250 mm×4.6 mm,5 μm),乙腈(A)-0.3%磷酸水溶液(B)为流动相进行梯度洗脱,柱温30 ℃,流速1 mL·min-1,检测波长254 nm,进样量10 μL,利用"中药色谱指纹图谱相似度评价系统2004 A版"对指纹图谱进行相似度评价。结果建立了化香树果序药材HPLC指纹图谱,所建立的化香树果序指纹图谱测定方法精密度、稳定性、重复性良好;通过12批样品指纹图谱的比较,确定了15个共有峰,指认了2个峰;样品中没食子酸和鞣花酸的含量较高,四川广元产地化香树果序没食子酸和鞣花酸含量最高。结论首次建立了化香树果序药材HPLC指纹图谱,为化香树果序药材的鉴定、含量测定、质量标准的建立提供参考依据。

Abstract:

Objective Simultaneous determination of gallic acid and ellagic acid content in Platycaryastrobilacea Sieb.et Zuccby HPLC method and fingerprint study on 12 batches of Platycaryastrobila cea Sieb.et Zucc.Methods The chromatographic fingerprints were built by using Acclaim TM 120A C18(250 mm×4.6 mm,5 μm)as stationary phase and acetonitrile(A)-0.3% phosphate aqueous solution(B)as mobile phase with gradient elution.The column temperature was maintained at 30 ℃,the flow rate at 1 mL·min-1,the injection volume at 10 μL and the detection wavelength was set at 254 nm.The similarity evaluation of the fingerprint was carried out by using 2004 A edition of the similarity evaluation system of traditional Chinese medicine chromatogram fingerprint.Results HPLC fingerprint was established,and the method was accurate,stable and repeatable;By comparing the fingerprints of 12 batches of samples,15 common peaks were selected and 2 peaks were identified;The contents of gallic acid andellagic acid were higher in the sample.The content of gallic acid and ellagic acid in Platycaryastrobilacea Sieb.et Zucc was the highest in Guangyuan area of Sichuan.Conclusion HPLC fingerprint was established for the first time to provide reference basis for identification,content determination and establishment of quality standard of Platycaryastrobilacea Sieb.et Zucc.
